基于乙醇胁迫酿酒酵母模型的抗衰老中草药筛选研究Screening of anti-aging Chinese herbal medicines based on ethanol stress model of Saccharomyces cerevisiae

摘要(Abstract):
酿酒酵母(Saccharomyces cerevisiae)在食品、化工、生物技术领域具有重要的工业应用价值,延缓其细胞衰老有助于提升生产效率和产品稳定性。本研究从28种中草药中筛选出具有延缓酿酒酵母衰老作用的品种。结果表明,加入质量浓度为1.0 mg/mL的丁香、车前草和连翘水提物,能够显著(P<0.05)促进酿酒酵母细胞的生长并延缓细胞衰老;通过流式细胞术结合碘化丙啶(PI)染色实验发现,这3种中草药的最适质量浓度为1.0 mg/mL;扫描电子显微镜(SEM)结果显示,衰老的酿酒酵母细胞表面粗糙出现明显褶皱,细胞之间出现黏连现象,加入中草药水提物后,大多数细胞表面变得光滑、形态饱满且完整。研究结果表明,丁香、车前草和连翘水提物能有效提升酿酒酵母的抗衰老能力。本研究为提升酿酒酵母在发酵后期的细胞活力、提高整体发酵效率提供了新思路。
